The Rare Mutation That Makes People Immune to Viruses

Researchers have discovered that a rare genetic mutation provides certain individuals with immunity to various viruses. This finding has led to the development of an experimental mRNA therapy aimed at replicating this effect, which showed success in preventing influenza and COVID-19 in animal trials. The study was published on August 13 in *Science Translational Medicine*.

Columbia University immunologist Dusan Bogunovic first identified this mutation, which causes a deficiency in an immune regulator called ISG15, around 15 years ago. Individuals with this mutation experience mild, persistent inflammation that appears to equip their bodies with enhanced antiviral capabilities. In the current project, Bogunovic’s team created a therapy that temporarily induces similar antiviral protection by promoting the production of 10 specific proteins associated with immunity.

In tests involving hamsters and mice, the therapy administered via a nasal drip effectively inhibited the replication of influenza and SARS-CoV-2, while also reducing illness severity. The research aims to create a drug that potentially offers broad antiviral protection, especially relevant for first responders and high-risk populations during future pandemics.

While the initial findings are promising, the therapy’s delivery and absorption must be refined before moving to human trials.
